#1 - Clinical and histological aspects of localized and generalized canine demodicosis

Abstract in English:

Demodicosis is a skin condition caused by Demodex mite, frequently diagnosed in clinical routine. The purpose of this study was to compare the clinical and histological manifestations of localized and generalized demodicosis in dogs, relating the amount of mites and microscopic lesions. The study group was formed by 46 dogs, 28 with generalized demodicosis, and 18 with the localized form of the disease, all of them with positive skin scrapings. Macroscopic lesions data in dermatological form were noted, and skin samples were collected for microscopic study. The histopathological analysis was individually performed by three reviewers without intercommunication, being considered the result that more occurred. Alopecia, scaling, erythema, and crusting were the most common clinical lesions in both forms of the disease. Histological severe dermatitis occurred especially in dogs with localized and generalized disease that had concomitant pyoderma and Demodex sp. mites were visualized with intensity moderate and severe in the most dogs with both forms of the diseases. Perifolliculitis was the most evident microscopic follicular change in this study, followed by folliculitis and furunculosis wall, which had higher relative occurrence in dogs with localized demodicosis. According to the results obtained, the severity of histologic lesions does not necessarily correspond to the clinical presentation of the disease, in addition to the amount of mites that apparently is not indicative of localized or generalized clinical lesions, and cannot differentiate clinical disease by histopathology.

Demodicose é uma afecção cutânea causada pelo ácaro Demodex sp. diagnosticada com frequência na rotina clínica veterinária. Objetivou-se descrever as manifestações clínicas e histológicas de cães com demodicose nas formas localizada e generalizada, relacionando a quantidade de ácaros e os achados microscópicos ao quadro clínico. Foram estudados 46 cães, sendo 28 com demodicose generalizada e 18 com a forma localizada da doença, dos quais todos possuíam raspados de pele positivo. Destes caninos foram anotados dados de lesões macroscópicas em ficha dermatológica e coletaram-se amostras de pele para estudo microscópico. A análise histológica foi realizada por três avaliadores e os dados registrados sem comunicação entre os mesmos, sendo considerado o resultado que mais ocorreu. Alopecia, descamação, eritema e crostas foram lesões clínicas comuns às duas formas de apresentação da doença que mais ocorreram. Inflamação histológica dermal grave ocorreu especialmente nos cães com doença localizada e generalizada que possuíam piodermite concomitante e ácaros Demodex sp. foram visualizados com intensidade entre moderada e acentuada na maioria dos cães com ambas as formas da enfermidade. Perifoliculite foi a alteração folicular microscópica mais evidenciada nos caninos deste estudo, seguida por foliculite mural e furunculose que tiveram maior ocorrência relativa nos cães com demodicose localizada. De acordo com os resultados obtidos verificou-se que a gravidade das lesões histológicas não corresponde necessariamente à forma de apresentação clínica da enfermidade, além disso, a quantidade de ácaros não demonstrou ser indicativa de lesões clínicas localizadas ou generalizadas, bem como não é possível diferenciar a doença clínica através do exame histopatológico.

Podocnemis expansa (Giant South American turtle) is the largest freshwater chelonian of South America. This turtle is a widely distributed specie, however its anatomical features are poorly described. The objective of this study was to describe the oropharyngeal cavity of the turtle. Twelve heads of 3 to 8-year-old female and male P. expansa turtles with an average body weight of 7.5kg were used. P. expansa has a sharp and well developed rhamphotheca, which together with a powerful jaw muscles and a large tongue uniformly distributed on the oropharynx floor works for the apprehension and swallowing of food, what ensures good adaptation in different environments.

Sendo assim, a caça, a pesca e a procura por ovos destes animais tem ocorrido frequentemente. Podocnemis expansa (Tartaruga da Amazônia) é o maior quelônio de água doce da América do Sul. É uma espécie largamente distribuída, porém, nota-se que aspectos anatômicos da espécie são insuficientemente descritos. O objetivo desse estudo foi descrever a cavidade orofaríngea desses quelônios. Foram utilizadas 12 cabeças de P. expansa adultas, de ambos os sexos, com idade entre 3 a 8 anos, com peso corpóreo de 5 a 22kg (média de 7,5kg). P. expansa conta com a presença de rafontecas afiadas e bem desenvolvidas que, associadas à musculatura potente da mandíbula e à língua volumosa e bem distribuída no assoalho da orofaringe, atuam na apreensão e deglutição do alimento, garantindo uma maior adaptação em diversos ambientes.

E-mail: rafalima@ufpa.br The agouti species Dasyprocta prymnolopha (D. prymnolopha) is a medium-sized rodent, diurnal, and characteristic of northeastern Brazil, south of the Amazon. Several studies have been made on these rodents. However, there is a lack of analysis of masticatory system, in particular morphology of the teeth. Thus, this research seeks to describe anatomical and histological aspects of the agouti teeth. For this purpose, we used adult agouti, in which measurements and descriptions of teeth and dental tissues were made. It was observed that the dental arch of D. prymnolopha comprises of twenty teeth, evenly distributed in the upper and lower arch, being inferior teeth larger than their corresponding higher. The incisors are larger, and between the posterior premolars and molars, there is a gradual increase in length in the anterior-posterior arch. In microscopic examination, a prismatic appearance was observed consisting of enamel prisms arranged in different directions, behind the enamel and dentin with standard tubular dentinal tubules with variable diameter and far between, also showing a sinuous path from the inner portion to the junction with more superficial enamel. Morphological analysis of dental tissues showed that an enamel with structural organization adapted to the act of chewing and high impact dentin compatible with standard tubular function resilience and mechanical damping of masticatory forces, as found in larger animals, confirming the understanding of eating habits that define much of its ecological functions within the ecosystem they inhabit.

Vários estudos têm sido feitos sobre estes roedores. No entanto, há uma carência de estudos do sistema estomatognático, em particular, a morfologia dos dentes. Assim, esta pesquisa procura descrever aspectos anatômicos e histológicos dos dentes cutia. Para isto, nós utilizamos cutias adultas, em que as mensurações e as descrições dos dentes e dos tecidos dentais foram feitas. Observou-se que a arcada dentária de D. prymnolopha é composta por vinte dentes, distribuídas uniformemente no arco superior e inferior, sendo os dentes inferiores, maiores do que os seus correspondentes superiores. Os incisivos são maiores, e entre os pré-molares e molares posteriores, existe um aumento gradual no comprimento do arco anterior-posterior. No exame microscópico, uma forma prismática foi observada o que consiste de prismas de esmalte dispostos em diferentes direções, atrás do esmalte e dentina com túbulos dentinários com padrão tubular de diâmetro variável e distantes entre si, mostrando também um caminho sinuoso a partir da parte interna da junção com o esmalte mais superficial. A análise morfológica dos tecidos dentários mostrou um esmalte com a organização estrutural adaptada para o ato de mastigar e dentina de alto impacto compatível com a função do padrão tubular de resiliência e amortecimento mecânico de forças mastigatórias, como encontrado em animais maiores, confirmando o entendimento de hábitos alimentares que definem muito das suas funções ecológicas dentro do ecossistema em que vivem.


#4 - Platelet activation: Ultrastructure and morphometry in platelet-rich plasma of horses, 32(1):83-92

E-mail: msouza@ufv.br This study was conducted to investigate the activation ability of the platelet-rich plasma (PRP) by pharmacological agents, as well as to verify the need or not of this activation for therapeutic use. The PRP was obtained from four healthy crossbred geldings aged 13 to 16 years (15±1years), and was processed for observation and quantification of the platelet morphology by using the transmission electron microscopy. All PRP samples were activated with 10% calcium chloride (CaCl2) solution, pure bovine thrombin or associated with CaCl2. The control (pure PRP) was not pharmacologically activated. In the pure PRP samples, 49% of the platelets were classified as state of activation uncertain, 41% as resting, 9% as fully activated and 1% as irreversibly damaged. Treatment with 10% CaCl2 provided a distribution of 54% platelets in state of activation uncertain, 24% as fully activated, 20% as resting, and 2% as irreversibly damaged. The platelet morphology of the bovine thrombin treated samples did not fit into classification adopted, as showing irregular shape with emission of large filamentous pseudopods, appearance of ruptured and whole granules in the remaining cytoplasm and extracellular environment. There was effect of the treatment on the platelet morphology (P=0.03). The 10% CaCl2 is an adequate platelet-activating agent. However, in cases the use of PRP under its liquid form is necessary, the use of pure PRP is recommended, since besides presenting an adequate percentage of fully activated platelets it also has significant amount of the resting type, which can be activated by substances found in the injured tissue.

O PRP foi obtido de quatro equinos mestiços hígidos, machos castrados, com 13 a 16 anos (15±1anos) de idade, e processado para observação e quantificação da morfologia plaquetária mediante a utilização da microscopia eletrônica de transmissão. Todas as amostras de PRP foram ativadas com cloreto de cálcio (CaCl2) a 10%, trombina bovina pura ou associada a CaCl2. O controle (PRP puro) não foi ativado farmacologicamente. Nas amostras de PRP puro, 49% das plaquetas foram classificadas como ativação incerta, 41% em repouso, 9% totalmente ativada e 1% com dano irreversível. O tratamento com CaCl2 a 10% proporcionou uma distribuição de 54% de plaquetas com ativação incerta, 24% totalmente ativada, 20% em repouso, e 2% como com dano irreversível. Amostras tratadas com trombina bovina apresentaram morfologia plaquetária que não se enquadraram na classificação adotada, apresentando forma irregular com emissão de grandes pseudópodes filamentosos, aspecto de rompimento e grânulos inteiros no citoplasma remanescente e meio extracelular. Houve efeito do tratamento sobre a morfologia plaquetária (P=0,03). O CaCl2 a 10% é um adequado agente ativador de plaquetas. Entretanto, nos casos onde se faz necessário o uso de PRP na forma mais líquida, recomenda-se o uso do PRP puro, que além de apresentar uma adequada porcentagem de plaquetas totalmente ativadas, também possui importante quantidade do tipo em repouso, que pode ser ativado por substâncias presentes no tecido lesionado.

E-mail: marcelloroza@gmail.com Eleven dogs and four cats with buccodental alterations, treated in the Centro Veterinário do Gama, in Brasilia, DF, Brazil, were submitted to cone beam computed tomography. The exams were carried out in a i-CAT tomograph, using for image acquisition six centimeters height, 40 seconds time, 0.2 voxel, 120 kilovolts and 46.72 milliamperes per second. The ideal positioning of the animal for the exam was also determined in this study and it proved to be fundamental for successful examination, which required a simple and safe anesthetic protocol due to the relatively short period of time necessary to obtain the images. Several alterations and diseases were identified with accurate imaging, demonstrating that cone beam computed tomography is a safe, accessible and feasible imaging method which could be included in the small animal dentistry routine diagnosis.

E-mail: marcelloroza@gmail.com Onze cães e quatro gatos, portadores de alterações buco-dentárias e atendidos no Centro Veterinário do Gama, em Brasília, DF, foram submetidos à tomografia computadorizada de feixe cônico. Os exames foram realizados em um tomógrafo i-CAT, utilizando para aquisição das imagens, altura de seis centímetros, tempo de 40 segundos, 0,2 voxel, 120 kilovolts e 46,72 miliampéres por segundo. O melhor posicionamento dos animais para realização do exame foi definido neste estudo. Esse é um fator fundamental para a realização do exame, que necessitou um protocolo anestésico simples e seguro, em função do tempo mínimo necessário à obtenção das imagens. Várias alterações e enfermidades foram identificadas, com extrema acurácia, credenciando a tomografia computadorizada de feixe cônico como um exame seguro, acessível e exeqüível e que pode ser incorporado à rotina odontológica das clínicas de pequenos animais.

Tendinitis of the SDFT was provoked in both forelimbs via intratendineous administration of 2.5mg of collagenase (2.5mg.mL-1), what was considered as beginning of the experimental phase. Twelve days after induced tendinitis, the horses were submitted to two treatments: (1) in the lesion caused in the right SDFT (treated group, TG), 2.5mL of PRP activated with calcium chloride at 0.0125mol.L-1, at concentrations from 320,000 to 500,000 platelets.µL-1, were administered; (2) in the tendinitis of the left SDFT (control group, CG), 2.5mL of 0.9% saline solution was administered. After 5 days, the animals were submitted to controlled and progressive physical activity during 30 days. Ultrasonographic examinations were carried out before and after tendinitis induction (48 hours after and on experimental days 7, 12, 14, 21 28, 35 and 42), with tendon cross-sectional area (CSA), lesion cross-sectional area (L-CSA), L-CSA percentage, severity and echogenicity of the lesion, as well as collagen fiber alignment being evaluated. The result revealed reduction (P<0.05) in the L-CSA and in the degree of lesion echogenicity in function of time, but with difference (P<0.05) between groups only for L-CSA, with smaller values in TG. The treatment with PRP promotes greater reduction in the area of the lesion measured by ultrasound.

#7 - Distrofia muscular nutricional em ovinos na Paraíba, p.120-124

[Nutritional muscular dystrophy in sheep in Paraíba.] Distrofia muscular nutricional em ovinos na Paraíba. Pesquisa Veterinária Brasileira 25(2):120-124. Centro de Saúde e Tecnologia Rural, UFCG, Campus de Patos, 58700-000 Patos, PB Brazil. E-mail: riet@cstr.ufcg.edu.br An outbreak of nutritional muscular dystrophy is reported in the semiarid region of northeastern Brazil affecting 3-4 months old Dorper sheep. The animals, weighing 30-40 kg, were fed ad libitum with milk, concentrated ration, Tifton hay, and a mineral mixture. Six out of 70 lambs were affected and died in the first 48 hours after the onset of the outbreak. Clinical signs were fever of 40-41ºC, incoordination followed by paralysis and recumbence, depression, prostration with decreased pupillary and corneal reflexes, decreased tonus of the tongue and maxilla, salivation, submaxillar edema, and increased cardiac and respiratory rates. Death occurred after a clinical manifestation period of 6-12 hours. At necropsy of three animals, skeletal muscles were pale, the liver was yellowish and enlarged, the parotid, submaxillary, retropharyngeal, prescapular and mediastinal lymph nodes were enlarged with red surface, and red areas were observed on the lung surface. On histology, segmental muscular necrosis was observed in all skeletal muscles examined. The liver had centrilobular fatty degeneration, and congestion was observed in the lung and lymph nodes. From 48-96 hours after the begin of the outbreak, another 3 animals were affected. They were treated with Vitamin A, E and D complex; two of them died and one survived. On the same farm, a flock of 20 Santa Inês sheep of the same age as the affected animals, fed with the same food, but no milk, and another 900 sheep of different ages were not affected. The over nutrition of sheep with fast growing rates, and the stress caused by two days of water restriction to improve milk consumption had been predisposing factors for the occurrence of the disease. It is also possible that some of the minerals supplemented interfered with selenium availability.

E-mail: riet@cstr.ufcg.edu.br An outbreak of nutritional muscular dystrophy is reported in the semiarid region of northeastern Brazil affecting 3-4 months old Dorper sheep. The animals, weighing 30-40 kg, were fed ad libitum with milk, concentrated ration, Tifton hay, and a mineral mixture. Six out of 70 lambs were affected and died in the first 48 hours after the onset of the outbreak. Clinical signs were fever of 40-41ºC, incoordination followed by paralysis and recumbence, depression, prostration with decreased pupillary and corneal reflexes, decreased tonus of the tongue and maxilla, salivation, submaxillar edema, and increased cardiac and respiratory rates. Death occurred after a clinical manifestation period of 6-12 hours. At necropsy of three animals, skeletal muscles were pale, the liver was yellowish and enlarged, the parotid, submaxillary, retropharyngeal, prescapular and mediastinal lymph nodes were enlarged with red surface, and red areas were observed on the lung surface. On histology, segmental muscular necrosis was observed in all skeletal muscles examined. The liver had centrilobular fatty degeneration, and congestion was observed in the lung and lymph nodes. From 48-96 hours after the begin of the outbreak, another 3 animals were affected. They were treated with Vitamin A, E and D complex; two of them died and one survived. On the same farm, a flock of 20 Santa Inês sheep of the same age as the affected animals, fed with the same food, but no milk, and another 900 sheep of different ages were not affected. The over nutrition of sheep with fast growing rates, and the stress caused by two days of water restriction to improve milk consumption had been predisposing factors for the occurrence of the disease. It is also possible that some of the minerals supplemented interfered with selenium availability.


#8 - Perinatal mortality of lambs in the county of Uruguaiana, Rio Grande do Sul

The following results reflect the lamb mortality rates of two neighbouring flocks of sheep in the district of Uruguaiana, Rio Grande do Sul. Post mortem examination was performed on 200 lambs, in order to establish the causa mortis and its relationship to sex, weight and climatic conditions. Perinatal mortality is a complex problem that does not have one specifc cause, but is the result of a variety of interrelating factors which are believed to differ from flock to flock. In this study, the non-infectious causes were found to be more important than the infectious ones. Based on the criteria used, the mortality was olassified as follows: complex of starvation/exposure to climatic conditions 157 (78.5%); dystocia, 21 (10.5%); predators, 9 (4.5%); stillbirth, 5 (2.5%); traumatism, 5 (2.5%); congenital defects, 1 (0.5%); neonatal infections, 1 (0.5%); without diagnosis, 1 (0.5%). There were a total of 119 males and 81 females. The mean weight of all lambs subrnitted to necropsy was 3,127 g, ranging from 1,400 to 5,100 g. The highest rate of mortality in relation tó weight was recorded in the group weighing between 3,000 and 3,500 g, 31 % of the total number of animals examined.

Estes resultados refletem a situação da mortalidade perinatal que ocorreu em dois rebanhos de propriedades vizinhas no município de Uruguaiana, Rio Grande do Sul, onde foram necropsiados 200 cordeiros no sentido de se tentar estabelecer o diagnóstico da causa mortis e, quando possível, correlacioná-la com o sexo, peso e as condições climáticas. A mortalidade perinatal de cordeiros é um problema complexo que não tem apenas uma causa específica, sendo, pois, o resultado do envolvimento de muitos fatores, os quais, acredita-se, se podem diversificar de rebanho para rebanho. Durante este estudo, as causas não infecciosas foram mais importantes que as infecciosas. Conforme os critérios adotados, a mortalidade ficou distribuída nos seguintes grupos: complexo da inanição/exposição às condições climáticas 157 (78,5%); distocia, 21 (10,5%); predadores, 9 (4,5%); natimortos, 5 (2,5%); traumatismo, 5 (2,5%); sem diagnóstico, 1 (0,5%). No total, foram 119 machos e 81 fêmeas. A média dos pesos de todos os cordeiros necropsiados foi de 3 .127 g, variando de 1.400 a 5.100 g. A maior freqüência de mortalidade em relação ao peso foi entre 3 .000 a 3.500 g, perfazendo 31 % dos animais examinados.
